灯盏花素对糖尿病大鼠肾脏蛋白激酶Cα和肿瘤坏死因子α表达的影响 被引量:6

Effects of breviscapine on expressions of PKCα,TNFα in kindey of diabetic rats

摘要 目的:探讨灯盏花素(breviscapine,Bre)对链脲佐菌素(STZ)诱导的糖尿病大鼠肾脏蛋白激酶Cα(PKCα)、肿瘤坏死因子α(TNFα)表达的影响。方法:采用SD大鼠尾静脉注射STZ45 mg.kg-1建立实验性糖尿病模型。将成模DM大鼠随机分成:糖尿病模型组(DM组)、灯盏花素治疗组(Bre-L组,10 mg.kg-1.d-1;Bre-H组,20 mg.kg-1.d-1),同时设正常对照组(NC组)。治疗8周后检测各组大鼠血糖(FBG)、肾指数、肌酐(SCr)、尿素氮(BUN)、尿白蛋白排泄率(UAER)、HE染色光镜观察肾脏形态结构,采用免疫组化方法检测肾脏PKCα、TNFα表达情况。结果:与DM组比较,Bre组大鼠血糖无明显变化,但肾指数、Scr、BUN、UAER均明显降低,光镜下肾脏结构亦明显改善,肾组织PKCα、TNFα的表达显著减少。结论:Bre对糖尿病大鼠肾脏具有一定的保护作用,可能与其抑制糖尿病大鼠肾脏PKCα、TNFα表达有关。 OBJECTIVE To investigate effects of breviscapine on renal function and expressions of PKCa ,TNFa in kindey of diabetic rats induced by STZ. METHODS 30 SD rats were used to create the experimental model of diabetes by injecting STZ through vena caudalis. They were randomly divided into untreated group(DM) and breviscapine groups(Bre). Another 10 rats were set as normal control group. Eight weeks later, blood glucose, renal index, SCr,BUN, UAER were assayed. The morphological change of renal cortex was observed by light microscope. Immunohistochemistry method was used to determine the expressions of PKCa and TNFa in renal tissue. RESULTS Compared with DM group, in Bre groups, the blood glucose level was unchanged, whereas renal index and UAER were decreased, and SCr, BUN were improved obviously. Kindey structure was improved obviously too. Moreover, expressions of PKCa and TNFa in renal tissue of diabetic rats in Bre groups were much less than those in DM group. CONCLUSION Breviscapine has protective effects on kindey in diabetic rats, which may be associated with lessening expressions of PKCa and TNFa in renal tissue.
