摘要
目的:研究异丙酚治疗早期脓毒症时肺水通道蛋白1(aquaporin 1,AQP1)基因表达的差异和对TNF-α的影响。方法:按完全随机设计,分为正常组(N组),脂多糖组(L组),脂多糖+异丙酚组(LP组),每组20只(n=20)。每组再根据注射药物后1、2、4、8h的不同时间点,随机分为T1、T2、T4、T8亚组(n=5)。各组于相应时间点处死动物,留取肺标本,以RT-PCR及相关软件分析各组AQP1 mRNA表达;用ELISA法测血清TNF-α浓度。结果:脂多糖对肺AQP1 mRNA的表达有明显的抑制作用,尤其在注射1h后表达明显减少(P<0.05),2~8h逐渐上升,但仍低于对照组(P<0.05);异丙酚能使脓毒症大鼠AQP1基因上调(P<0.05),在早期(1h)差异有统计学意义(P<0.05)。血清TNF-α浓度,L组注射LPS后1hTNF-α血清水平骤然升高至峰值(P<0.05),至4h后渐趋平稳,但各时间点浓度水平均高于对照组(P<0.05),异丙酚治疗组各时间点TNF-α值较L组明显降低(P<0.05),于1h后渐升高,于4h达到峰值(P<0.05)。结论:注射LPS1h后可迅速下调肺组织AQP1基因的表达和使TNF-α血清水平骤然升高至峰值;异丙酚治疗脓毒症大鼠后,早期(1h后)肺组织AQP1mRNA表达明显上调,降低了TNF-α浓度。
Objective To observe Aquaporin 1 (AQP1) gene expression of lung and TNF-α in serum in early septic rats. Methods Rats were randomly divided into Saline group (N), Lipopolysaccharides group (L), Lipopolysaecharides + propofol group (LP group). Then each group were divided randomly into four sub-groups of five samples (n = 5): T1, T2, T4, T8, after injection in 1, 2, 4, 8 h. which were sacrificed at corresponding time points to analyze AQP1 mRNA expression with lung specimens by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR), and measure the concentrations of TNF-a in serum by ELISA. Results The expression of AQP1 mRNA were inhibited significantly by LPS, especially in lh after injection (P 〈 0.05), then gradually increased in 2-8 h, but still lower than the control group (aP 〈 0.05 ). Propofol increased the expression of AQP1 mRNA (P 〈 0.05), especially in 1 h. The concentrations of TNF-α rose to the peak after injection of LPS with L group in I h (P 〈 0.05), then into the stabilizition state in 4 h, but the average value was higher thancontrol group at each time(P 〈 0.05). In LP, the value of TNF-α was significantly lower than L at each time point (P 〈 0.05), which increased gradually after 1 h, then rose to the peak in 4 h (P 〈 0.05). Conclusion The expression of AQP1 mRNA was reduced after the injection of LPS in lh, while the TNF-α in serum increased to the peak. The expression of AQP1 mRNA was significantly increased in the early period (1 h later), and the concentration of TNF-α in serum was reduced in LP, and delayed its peak level to 4 h.
