摘要
[目的]对CO2超临界萃取的厚朴提取物的HPLC指纹图谱进行研究。[方法]采用高效液相色谱;PhenomenexLuna 5u C18(2)100R(4.6 mm×250 mm,5μm)为色谱柱;以无水甲醇-浓度0.1%醋酸(74∶26,V/V)梯度洗脱,流速为1.0 ml/min;检测波长为280 nm;柱温35℃。[结果]在重现性试验中,图谱相似性均大于0.99,所以HPLC特征指纹图谱可作为控制CO2超临界厚朴提取物的质量标准;10批厚朴提取物相似度在0.9~1.0之间,表明厚朴SFE-CO2提取物质量均一、稳定。[结论]厚朴标准提取物的纯度非常高,主要有效成分厚朴酚和和厚朴酚的含量几乎达到100%,杂质成分的含量非常少。
[ Objective ] To study HPLC fingerprints of CORTEX MAGNOLIAE OFFICINAH$ extracts from supereritical CO2 extraction, and to provide effective methods for quality control. [ Method ] A HPLC analysis was performed at 35 ℃on a chromatographic column Phenomenex Luna 5u Cl8 (2) 100R (4.6 mmx 250 turn ,5 pma) with the detection wavelength at 280 nm. The mobile phase consisted of anhydrous metha- nol and 0. 1% acetic acid (WV,74:26). Following the injection of sample solution,an elution was initiated at a flow rate of 1.0 ml/min. [ Re- sult] In repeatability test ,the fingerprint similarity among samples was all greater than O. 99, so that HPLC-fingerprints can be use for quality control of CORTEX MAGNOLIAE OFFICINALIS extracts frem supereritical CO2 extraction. The similarity among 10 extracts ranged from 0.9 to 1. O,which showed that SFE-CO2 extracts were uniform and stable. [ Conclusion] CORTEX MAGNOLIAE OFFICINALIS extracts were proved to be high-ouritv oroducts with very low imouritv .in which main active ingredients magnolol and honokio could nearly occupy 100%.
