发育早期双酚A暴露对成年期雌性大鼠血糖及脂联素水平的影响 被引量:4

Effect of Exposure to Bisphenol A during Early Development on Levels of Serum Glucose and Adiponectin in Female Rats

摘要 目的研究发育早期暴露双酚A(BPA)对成年期雌鼠血糖及血清脂联素水平的影响。方法将在2周内妊娠的母鼠随机分成3组,分别为对照(含1%无水乙醇的水)组、低剂量(1 mg/L)BPA染毒组和高剂量(10 mg/L)BPA染毒组,每组4只,单笼饲养。从妊娠第6 d至哺乳期结束,采用自由饮水方式进行染毒。测量雌性仔鼠空腹血糖、胰岛素及脂联素水平,计算胰岛素抵抗指数。结果与对照组比较,各剂量BPA染毒组子代雌鼠哺乳期及断乳期体重均显著增高(P<0.05),空腹血糖和胰岛素水平也显著增高(P<0.05),血清脂联素水平显著降低(P<0.05)。但低、高剂量BPA染毒组仔鼠哺乳期及断乳期体重,空腹血糖、胰岛素、血清脂联素水平和胰岛素抵抗指数间比较,差异均无统计学意义。结论发育早期暴露双酚A可诱导成年期子代雌鼠肥胖,引起胰岛素抵抗;血清脂联素水平的降低可能与胰岛素抵抗的发生有关。 Objective To study of bisphenol A (BPA) exposure during early development on the levels of serum glucose and adiponeetin in female rats. Methods The pregnant rats of two weeks old were randomly divided into three groups ,four in each,the control group (1% of anhydrous alcohol with water),low dose (1 mg/L) and high doses (10 mg/L) groups of BPA exposed through drinking water,from gestation day 6 to the end of lactation respectively.Eighty days after weaning, the change of body weight and the levels of blood glucose, insulin and adiponectin were determined. The insulin resistance index in female offspring was calculated. Results Compared with controls,the body weight was significantly higher in BPA-exposed female offspring (P〈0.05), the levels of blood glucose and insulin increased significantly, and the level of serum adiponectin decreased significantly. Conclusion BPA exposure during early development may induce obesity in aduhhood,and result in insulin resistance. The decreased level of serum adiponectin maybe is associated with insulin resistance.
