摘要
目的探索建立一套无创、廉价、快速的小鼠气管插管、滴注方法。方法建立一套小鼠透射灯照射下经口咽气管插管、滴注的方法,并通过对28只雄性C57BL/6J小鼠(体重20—30g)以1次/2d的频率重复插管、滴注0.06ml炭黑颗粒物(2.5mg/m1)共6次来测试该方法的可行性。结果共成功完成162次插管、滴注操作,末次滴注后解剖小鼠可见双肺散在分布炭黑颗粒物。结论透射灯下经口直视气管滴注法具有无创、价廉、快速等特点,可广泛用于小鼠呼吸系统重复滴注。
Objective To develop an inexpensive rapid method for repeated endotracheal intubation and instillation in mice. Methods A set of special operations to facilitate oropharyngeal exposure were described, such as the transillumination of the neck to facilitate visualization of the trachea through the oropharynx, readily available polyethylene cater to intubate the trachea, and a simple method to control the operation quality. Twenty-eight male C57BL/6J mice (between 20 and 30 g) were repeatedly intra-traeheally instilled with 0.06 ml ultra-fine carbon black (ufCB, 2.5 mg/ml) for 6 times (once per 2 days). Results In all, the instillation-operations were conducted successfully for 162 times. The mice were sacrificed after the 6th repeated instillation and the dispersion of the ufCB was found in both the left and right lungs. Conclusion The improved directviewing intubation method through oropharynx under the trans-illumination features the non-invasive inexpensive and rapid characteristics, and can be widely used in mice endotracheal instillation.
