血红素加氧酶在局灶性脑缺血再灌注大鼠大脑组织表达测定

摘要 [目的]测定血红素加氧酶(HO)在局灶性脑缺血再灌注大鼠大脑组织表达。[方法]使用随机平行对照方法,将SPF级雄性大鼠20只随机分为两组,缺血组15只参照Zea-logue方法造大鼠大脑中动脉缺血2h再灌注24h模型,假手术对照组5只采用同样方法暴露颈动脉后缝合切口。观察脑组织形态结构,测定HO-1表达。[结果]假手术组脑组织形态结构正常,模型组大鼠大脑缺血区组织明显水肿。HO-1表达相同部位缺血组明显高于假手术组(P<0.01)。[结论]局灶性脑缺血再灌注大鼠大脑血HO升高,HO-1参与了局灶性脑缺血再灌注损伤。 [ Objective ] To investigate the effects of the expression of HO - 1 in cerebral tissues of rat after focal cerebral ischemia and reperfusion. [ Methods ] 2 hours right intraluminal middle cerebral artery occlusion with reperfusion was induced in 20 rats,and the specimen of brain were performed at the point after 24 hoursreperfusion ;expressions of HO -1were detected by genechip profile and RT- PCR method. [ Results] Expres- sions of HO - 1 were seen sparsely in the control specimens, and were markly increased afer transient focal cerebal ischemia and reperfusion(P 〈0.01 ). [ Conclusion] Expression of HO- 1 is a significant response totransient focal cerebral ischemia/reperfusion and contributes to the early formasion of vasogenic edema that can be observed all parts of brains.
