两种胶体金制备方法的比较研究 被引量:3

摘要 通过控制还原剂的加入量(20 mL 0.01%四氯金酸溶液中1%柠檬酸钠和抗坏血酸添加量分别为300μL与400μL),利用柠檬酸钠和抗坏血酸还原两种方法制备得到了平均粒径为20 nm的胶体金颗粒,并对其进行了抗体标记比较研究.结果显示:抗坏血酸还原法制备得到的胶体金标记的蛋白量为15.18μg/mL(蛋白质量/胶体金体积,下同),高于柠檬酸钠还原法的5.07μg/mL,间接ELISA法测得其效价(1∶800)也比柠檬酸钠还原法制备的颗粒标记物高(1∶400),这表明抗坏血酸还原法更适合于标记用胶体金颗粒的制备. In this paper, colloidal gold particles with the average particle size of 20 nm were prepared by sodium citrate reduction method and ascorbic acid reduction method by controlling the addition amount of reductants, and we also compared the antibody labeling. The results showed that the amount of protein labeled by colloidal gold prepared by ascorbic acid reduction method was 15.18μg/mL, which was higher than that of the sodium citrate reduction method (5.07 μg/mL); and the titer of the former determined by intermediate ELISA was 1:800, which was higher than that of the latter (1:400). The results indicated that ascorbic acid reduction method was more suitable for the preparation of colloidal gold particles for labeling in comparison with the sodium citrate reduction method.
