摘要
为了节省IHA(间接血凝试验)血凝抗原,扩大家畜O型口蹄疫免疫抗体检测数量,在不影响检测效果(合格率)的基础上,探索和改进原正向间接血凝试验检测方法。试验分别用口蹄疫O型-Asia 1型二价灭活苗和猪O型口蹄疫苗对天水地区几个养殖场的家畜进行了免疫注射,免疫后(牛免疫21 d、猪免疫28 d)随机抽样采血,常规分离血清,采用简化后的正向间接血凝试验进行牛和猪口蹄疫O型免疫抗体检测试验。结果表明:免疫合格率分别达到97.5%和92.5%;改进后的IHA既加快了操作速度、节省诊断抗原,又降低了检测成本。在家畜O型口蹄疫检测工作中具有推广应用价值。
To save Indirect Hemagglutination Test(IHA) hemagglutinin antigen and increase the number of detection of immune antibody against type O Foot-and-mouth disease virus,this study aimed to explore and improve IHA method,besed on without affecting the test results(pass rate).The livestock of the several farms in Tianshui area were vaccinated respectively with FMDV Type O and Asia1 Inactivated Vaccine and FMDV Type O in swine.After immunization(cattle for 21 d,swine for 28 d),tested the serum from random samples of blood by simplified-indirect and positive hemagglutination.The results indicated that the qualified immune rate was up to 97.5% and 92.5% respectively in the cattle and swine.The improved IHA method enhanced the operation speed,saved diagnostic antigenand and reduced the test cost.It has popularizing value in detection of livestock type O Foot-and-mouth disease.
