摘要
目的观察热疗在阿霉素对人急性髓系白血病细胞株KG-1a细胞杀伤作用中的増敏作用。方法四甲基偶氮唑蓝(MTT)法确定阿霉素的工作浓度,以该浓度进行化学治疗和热疗联合化学治疗,选择温度40、42℃,体外作用于KG-1a细胞。作用前及作用48 h后,采用台盼蓝拒染法检测肿瘤细胞的存活率;MTT法检测对肿瘤细胞增殖的抑制作用;流式细胞仪检测肿瘤细胞凋亡率的变化。结果各处理组的存活率明显要低于对照组(P<0.01),40、42℃热疗联合化学治疗组的细胞存活率明显低于单纯的热疗组(P<0.01)和化学治疗组(P<0.01)。40、42℃热疗联合化学治疗组的细胞抑制率明显高于化学治疗组(P<0.01)和单纯的热疗组(P<0.01)。各处理组的细胞凋亡率明显高于对照组(P<0.01),40、42℃热疗联合化学治疗组细胞凋亡率明显高于化学治疗组(P<0.01)和单纯的热疗组(P<0.01)。结论热疗可以增加阿霉素对KG-1a细胞的杀伤作用,提高肿瘤细胞的凋亡率。
Objective To observe the sensitization of hyperthermia to the adriamycin cytotoxicity in human KG-1a leukemic cell line. Methods The working concentration of adriamycin was determined by 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) assay.Chemotherapy and hyperthermia combined with chemotherapy were acted on KG-1a cell in vitro with the working concentration at 40 ℃ and 42 ℃.The survival rates of cancer cell were determined before treatment and 48 hours after the treatment by trypan bule exclusion test.The cell growth inhibition action was evaluated with MTT assay,and the apoptosis rate of KG-1a cell was determined by flow cytometric analyses. Results The survival rates in each treatment group were lower than those in control group(P0.01),and those in 40 ℃ and 42 ℃ hyperthermia combined with chemotherapy groups were lower than those in simple hyperthermia group(P0.01) and chemotherapy group(P0.01).The cell growth inhibition rates in 40 ℃ and 42 ℃ hyperthermia combined with chemotherapy groups were higher than those in simple hyperthermia group(P0.01) and chemotherapy group(P0.01).The apoptosis rate of cell in each treatment groups were higher than those in control group(P0.01),and those in 40 ℃ and 42 ℃ hyperthermia combined with chemotherapy groups were higher than those in simple hyperthermia group(P0.01) and chemotherapy group(P0.01). Conclusion Hyperthermia may increase the killing effect of adriamycin on KG-1a cell,and increase the apoptosis rate of tumor cell.
