血塞通致RBL-2H3细胞与原代肥大细胞脱颗粒特性的比较 被引量:7

摘要 【目的】通过比较血塞通注射液等在类过敏试验中致RBL-2H3细胞与大鼠腹腔原代肥大细胞脱颗粒特性的异同,以评价2种细胞在中药注射液类过敏试验中的意义。【方法】选用已经明确能引起类过敏反应的3种供试品即血塞通注射液、Compound 48/80和吐温80,分别与RBL-2H3细胞和大鼠腹腔肥大细胞共同孵育1 h,通过β-氨基己糖苷酶释放率和组胺释放率、释放量等的检测,分析比较脱颗粒特性的异同,同时观察了不同浓度血塞通注射液对脱颗粒程度的影响。【结果】2种细胞在血塞通注射液、Compound 48/80和吐温80的直接刺激下均出现脱颗粒现象,脱颗粒程度与血塞通注射液呈剂量依赖性;原代肥大细胞β-氨基己糖苷酶释放率、组胺释放量和释放率均显著高于RBL-2H3细胞,2种细胞比较差异有统计学意义(P<0.01)。【结论】在类过敏反应脱颗粒试验中,RBL-2H3细胞和原代大鼠腹腔肥大细胞脱颗粒具有相同趋势,均可用于中药注射液类过敏评价,RBL-2H3细胞应用方便,而肥大细胞更加敏感。 Objective To investigate the similarities and differences of degranulation in anaphylactoid reactions induced by Xuesaitong Injection (XI) between RBL-2H3 cells and rat peritoneal primary mast cells, and to evaluate the significance of RBL2H3 cells and rat peritoneal primary mast cells in anaphylactoid test. Methods RBL2H3 as well as rat peritoneal mast ceils were incubated with XI, Compound 48/80 and Tween-80 for one hour. And then we measured β-hexosaminidase release rate, histamine release rate and their release content for the analysis of the differences of degranulation between RBL2H3 cells and rat peritoneal primary mast cells. We also investigated the effect of different concentrations of XI on degranulation. Results The two kinds of cells showed degranulation after direct stimulation of XI, Compound 48 / 80 and Tween 80. And the degree of degranulation in the two kinds of cells was dose-dependent after treatment with different concentrations of XI. Primary mast cells presented higher β-hexosaminidase release rate, histamine release rate and histamine release content than RBL- 2H3 cells, and the differences were significant ( P 〈 0. 01 ). Conclusion RBL-2H3 cells and mast cells have the same trend of degranulation in the anaphylactoid reaction. Both of them can be used to evaluate the anaphylactoid reaction induced by traditional Chinese medicine injection, and RBL-2H3 cells are of convenient application while Drimarv mast cells have higher sensitivitv.
