摘要
目的从微观与宏观两方面同时探讨肌源干细胞(muscle-derived stem cells,MDSC)对肌肉内脂肪移植的辅助异化作用。方法实验共分A、B、C三组:A组为实验组:用MDSC细胞悬液与自体脂肪组织联合移植在小鼠的右侧后肢。B组为对照组:用PBS液取代了细胞悬液植入在左侧后肢。C组为健康雄性成年C57小鼠(15~20 g)的胫前肌作为空白对照组。通过大体观察、MRI成像、组织学染色比较各组之间的差异。结果大体观察治疗组小鼠后肢体积明显大于对照组;MRI成像发现MDSC治疗组的脂肪移植物显影显著强于对照组;组织学染色进一步发现MDSC治疗组多数由细胞结构完整,细胞形态良好的脂肪细胞组成。与之相反,对照组组织学染色可见广泛的纤维增生和大量的脂肪液化坏死。毛细血管计数结果显示,MDSC治疗组含有的毛细血管数量约是对照组的3倍。结论 MDSC与自体脂肪组织联合移植可以通过促进毛细血管新生而达到提高肌肉内脂肪移植物的长期存活率。
Objective To explore macroscopically and microscopically the auxiliary alienation effect of the MDSCs co-transplanted fat placed intramuscularly. Methods There were A, B and C three groups. Group A (experiment group) :MDSCs suspension and autologous fat tis- sues were co-transplanted into fight hind limbs of the recipient mice. Group B (control group) : PBS was used to replace the cell suspension and contralateral hind limbs served as the control side. Group C, tibialis anterior muscles from health male adult C57BL/6 mice (15 ~20 g) were used as blank control. The general observation, MRI scanning and histological staining among groups were compared. Results The volume of the mice hind limbs of experiment group was obviously larger than that of the control groups through the general observation. The MRI scanning showed stronger fat signal of MDSCs-treated group than that of control group. Moreover, histological evaluation exhibited well preserved and intact fall cells in MDSCs-treated group. In contrast, the control group showed extensive fibrosis and fat graft lost. Furthermore, the MDSCs-treated group possessed almost 3-fold of the capillary density to that of the control group. Conclusion Co-transplantation of muscle-derived stem cells and autologous fat tissuesimproves the long-term survival of intramuscular fat transplants through promoting neovascular- ization.
