葛根素对火药烟雾致支气管上皮细胞损伤的防护效应 被引量:3

摘要 目的研究火药烟雾损伤支气管上皮细胞模型中炎性细胞因子IL-8和IL-6的变化,以及葛根素对上述损伤的防护作用。方法用不同浓度(100、50、25、12.5μg/ml)的葛根素预处理细胞后,建立火药烟雾损伤支气管上皮细胞(BEAS-2B)模型。采用MTT法检测火药烟雾对细胞存活率的影响,ELISA法检测细胞培养上清液中IL-8和IL-6的浓度。结果以正常对照组细胞存活率作为100%,火药烟雾刺激后,BEAS-2B细胞的存活率下降至48.53%±4.44%,加入葛根素后细胞存活率明显提高,且呈剂量依赖性;细胞培养上清液中IL-8(P<0.05)、IL-6(P<0.01)浓度升高。加入葛根素后,IL-8、IL-6浓度逐渐下降(P<0.05),亦呈浓度依赖性。结论火药烟雾可致体外培养支气管上皮细胞存活率降低并促进炎性细胞因子IL-8和IL-6分泌,中药葛根素可逆转这种损伤效应。 Objective The present study investigates the secretion of inflammatory cytokines IL-8 and IL-6 by bronchial epithelial cells(BEAS-2B) injured by black powder smoke and the curative effect of puerarin.Methods BEAS-2B were assigned to pre-treat with puerarin at doses of 100,50,25,12.5μg/ml respectively,followed by irritation by black powder smoke.MTT method was used to observe the effect of the smoke(irritated for 10 minutes) on the cells.The IL-8 and IL-6 contents in the cell culture supernatant were detected using the ELISA method.Results After the cells were irritated with black powder smoke,their survival rate decreased(48.53%±4.44% of control),whereas the content of IL-8(P〈0.05) and IL-6(P〈0.01) in the cell culture supernatant increased.However,puerarin increased the survival rate of BEAS-2B after irritation and effectively inhibited the secretion of IL-8 and IL-6.Furthermore,the results show that the inhibitory effects were concentration dependent,and had similar statistical differences of P〈0.01.Conclusions Black powder smoke can damage BEAS-2B in vitro and induce the secretion of inflammatory cytokines IL-8 and IL-6.However,the Chinese herb medicine puerarin can reverse this injury and reduce the amounts of IL-8 and IL-6 secreted by the injured cells.
