摘要
目的 Dishevelled(Dvl)亚家族成员在非小细胞肺癌中均存在过表达,但他们与β-catenin和p120ctn及患者预后的关系尚不清楚。方法采用S-P免疫组化和RT-PCR法检测80例非小细胞肺癌(NSCLC)组织中Dvl-1、Dvl-3与β-catenin、p120ctn表达的关系及对预后的影响。结果正常支气管黏膜上皮Dvl-1、3均呈阴性表达,而在肺癌组织中表达上调,其异常表达率分别为52.5%(42/80)和51.3%(41/80),肺腺癌的阳性率均高于鳞癌,并与NSCLC的低分化高分期和淋巴结转移有关(<0.05)。Dvl-1高表达与β-catenin异常表达相关(<0.01),而Dvl-3高表达与p120ctn异常表达相关(<0.01)。Dvl-1、3的异常表达与非小细胞肺癌患者不良预后呈正相关(<0.05)。结论 NSCLC组织中存在着Dvl-1和Dvl-3的过表达,并与组织学分型、分化、临床分期和淋巴结转移相关。Dvl-1、3过表达分别与β-catenin、p120ctn异常表达相关。Dvl-1、3在非小细胞肺癌组织的异常表达可作为评估预后的辅助指标。
Objective Dishevelled(Dvl) family proteins exist and over-express in non-small cell lung cancer(NSCLC),but their correlation with the prognosis of patients is not clear.Methods We used immunohistochemistry and RT-PCR to assess the presence of Dvl-1,Dvl-3,β-catenin and p120ctn expression and their relationship with prognosis in 80 cases of non-small cell lung cancer specimens.Results Dvl-1 and Dvl-3 expressions in normal bronchial epithelium were negative,while the positive expression rate in primary tumors was 52.5%(42/80)for Dvl-1and 51.3%(41/80) for Dvl-3,with significantly higher in adenocarcinomas than in squamous cell carcinomas,and associated with poor tumor differentiation,high TNM stage and lymphnode metastasis(0.05).Dvl-1 positive expression was related to abnormal expression of β-catenin(0.01),while the Dvl-3 to that of p120ctn(0.01).The positive expression of Dvl-1,3 in non-small cell lung cancer was related to the poor prognosis of patients(0.05).Conclusion Overexpressed Dvl-1 and Dvl-3 in NSCLC are related to the tumor histological types,differentiation,clinical stage and lymphnode metastasis.Dvl-1,3 positive expression is related to the abnormal expression of β-catenin and p120ctn respectively,and also to poor prognosis in NSCLC.
