摘要
目的设计一种可提高组织切片苏木精-伊红(HE)染色速率及染色质量的病理组织切片染色装置。方法设计制作由水浴箱、带盖染色缸、电控箱三部分构成的组织切片恒温染色台,实验比对组织石蜡切片上的石蜡膜溶于二甲苯的互溶速率和镜下染色效果。结果在37.0℃条件下,石蜡膜在二甲苯中的溶脱时间比室温条件下缩短了近一半的时间,互溶速率两组比较差异有显著统计学意义(P<0.001);在减少1/2染色时间染制的切片组织色泽与室温条件下的染色效果类似,而且细小颗粒着色清晰。结论应用该设备条件的组织切片染色效果优于传统常温方法。
Objective To design a pathological tissue section staining apparatus, and improve the speed and quality of hematoxylin-eosin staining of tissue slices. Methods A constant temperature staining machine for histological section was designed and produced, which was composed of 3 parts that included water bath box, staining jars with covers and electric control box. Then the miscible rate of paraffin film in xylene and staining effect on paraffin-embedded tissue sections were compared. Results The striping time of paraffin film in xylene was reduced by nearly half at 37℃ than that at room temperature, and there was a statistically significant difference between the 2 groups(P 〈 0.001). The staining effect on tissue slices at 37 ℃ with 112 routine time was comparable with stained traditional procedure, and the tiny particles was clearly observed. Conclusion It is demonstrated the tissue section staining effect by the equipment is better than that by the traditional method at room temperature.
