摘要
目的探讨苍白球超级化激活环核苷酸门控阳离子(HCN)通道对氟哌啶醇所致帕金森病大鼠僵直症状的影响。方法大鼠单侧苍白球埋置套管,恢复3d后腹腔注射氟哌啶醇5mg/kg,在苍白球内微量注射生理盐水(生理盐水对照组)以及HCN通道特异性阻断剂ZD7288(0.5mmol/L,实验组),观察其对氟哌啶醇导致的大鼠僵直症状的影响。结果生理盐水对照组大鼠无明显偏转行为;实验组9只大鼠中有4只表现出明显的对侧偏转行为,大鼠头部-背部直线与背部-尾部直线所呈夹角评分与生理盐水对照组比较差异有显著性(u=132.500,P<0.001);另外5只表现为明显的同侧偏转行为,其评分与生理盐水对照组比较差异有统计学意义(u=9.000,P<0.001)。结论 HCN通道可能通过对苍白球神经元兴奋性的不同作用诱发大鼠出现不同的偏转行为。
Objective To investigate the effects of hyperpolarization activated cyclic-nucleotide gated(HCN) channel in globus pallidus on haloperidol-induced catalepsy in rats.Methods A cannula was implanted into unilateral globus pallidus of rats.After three days of recovery,5 mg/kg of haloperidol were intraperitoneally injected,and microinjection of normal saline(normal saline group) and 0.5 mmol/L of ZD7288(a specific blocking agent of HCN channels,experimental group) and its influence on stiffness symptoms caused by haloperidol in rats was recorded.Results The rats in normal saline group did not show obvious deflection,those in experimental group showed different extents of deflection,four of the nine rats manifested contralateral deviation,compared with normal saline group,the difference was significant(u=132.500,P〈0.001),the other five displayed significant ipsilateral deviation,the score was significant as compared with normal saline group(u=9.000,P〈0.001).ConclusionHCN channels induce various deflection in rats is probably through different effects of neuronal excitability of globus pallidus.
