摘要 目的研究大孔树脂分离纯化独一味总黄酮的工艺条件与技术参数。方法采用高效液相色谱法测定山栀苷甲酯和8-O-乙酰山栀苷甲酯的含量,紫外分光光度法测定独一味中总黄酮的含量。结果 D-101树脂对独一味总黄酮的富集效果最好。吸附条件为药液质量浓度1 g/mL,原液室温上样,吸附流速3 BV/h,洗脱剂为70%乙醇,洗脱流速5 BV/h,洗脱剂用量为10 BV。结论试验工艺稳定、可行。 Objective To research the separation and purification of Lamiophlomis rotata total flavonoids conditions and technical parameters. Methods Determination of gardenia glycoside 8- O- methyl and acetyl methyl glucoside by HPLC, single blind UV spec-trophotometry of total flavonoids in the investigated macroporous resin purification process. Results D- 101 resin for the enrichment of total flavonoids Lamiophlomis rotata best. Adsorption conditions were liquid concentration l g/mL (the equivalent of raw medicinal herbs) ,the sample liquid at room temperature,the adsorption velocity 3 BV/h(BV muhiple of the column bed volume),70% ethanol eluant,elution flow rate 5 BV/h,eluting agent is 10 BV. Conclusion The process is stable and usable.
