田口试验设计法优选灵芝多糖脂质体的制备工艺

摘要 目的优选灵芝多糖脂质体的最佳工艺。方法采用薄膜分散-微孔滤膜挤压法制备灵芝多糖脂质体,运用田口试验设计法以载药量的信噪比(S/N)作为指标,对脂药比、膜材比和超声时间等制备条件进行优选。结果薄膜分散-微孔滤膜挤压法制备灵芝多糖脂质体的最佳工艺为脂药比10:1,膜材比8:1,超声时间20min,水化温度为50℃。结论采用最佳制备工艺制作的灵芝多糖脂质体载药量较高。 Objective To optimize the preparation conditions of Ganoderma lucidum polysaccharides hposome (GLPS). Methods GLPS liposome was prepared by membrane distribution-micromembrane extruding method. Taguchi Methodology was adopted to optimize the preparation conditions which was designed with lecithin to drug ratio, lecithin to cholesterol ratio, and ultrasonic time, and the indexes of the drug-loading fate's Signal-to-noise ratio (S/N). Results The optimized preparation conditions were as follows: the ratio of lecithin to drug was 10:1, the ratio of lecithin to cholesterol was 8:1, the ultrasonic time was 20 min, the hydration temperature was 50"℃ Conclusion The GLPS liposome prepared under the optimized conditions has high drug-loading rate.
