摘要
甲烷氧化菌素是甲烷氧化菌分泌的能够捕获铜的一种生物螯合剂。研究了Cu2+在甲烷、甲醇作为碳源时,对甲烷氧化菌细胞浓度和甲烷氧化菌素发酵产量的影响;采用大孔树脂法及大孔树脂—固相萃取混合法纯化甲烷氧化菌素,探索固相萃取的条件,并通过HPLC分析其样品的纯度。结果表明,随着Cu2+浓度的增加,甲烷氧化菌细胞的生长速率有很大的提高,而甲烷氧化菌素的产量降低。大孔树脂和固相萃取法能够纯化甲烷氧化菌素。并且固相萃取法的洗脱体系为甲醇—水,洗脱梯度为60%。
Methanobactin,produced by Methanotrophs,is a kind of copper-binding biological chelating agent.This paper studies the effect of copper on cell concentration and methanobactin fermentation with methane and methanol as carbon source, using Diaion HP-20 column and uniting Diaion HP-20 column and C 18 solid-phase extraction(SPE) purities methanobactin, explores SPE conditions and analyzes the sample purity by HPLC.The results show that with the increase of Cu 2+ concentration, Methanotrophs growth rates of cells have greatly increased,and methanobactin has lower yields.Uniting Diaion HP-20 column and SPE can purity methanobactin and the elution system is methanol-water and elution gradient is 60% of SPE.
