骨形态发生蛋白-7调节肾病鼠Wnt1/β-catenin信号通路

Bone Morphogenetic Protein-7 Regulates Wnt1/β-catenin Signaling Pathway in Nephrotic Rats

摘要 目的观察骨形态发生蛋白-7(BMP-7)对嘌呤霉素氨基核苷酸肾病(PAN)鼠肾小球Wnt/β-catenin表达的影响,探讨BMP-7的肾保护作用及可能机制。方法 20只嘌呤霉素氨基核苷酸诱导的大鼠随机分为PAN组和PAN+BMP-7组(BMP-7组),每组各10只。BMP-7组每周2次腹腔注射人重组BMP-7 30微克/(千克.次)。另设正常对照组(NC,n=10)。第12周检测尿液红细胞(UE)、24h尿蛋白(24h UP),血清半胱氨酸蛋白酶抑制剂(Cys C)、巨噬细胞移动抑制因子(MIF)。处死大鼠,反转录-聚合酶链反应检测Wnt1和β-catenin mRNA表达水平。Western blotting检测Wnt1和β-catenin蛋白表达水平。结果与NC组相比,PAN组UE、24h UP、Cys C、MIC、肾小球Wnt1和β-catenin mRNA和蛋白质的表达水平均显著升高,差异具有显著的统计学意义(P均<0.01)。BMP-6组UE、24h UP、Cys C、MIF、肾小球Wnt1和β-catenin mRNA和蛋白质的表达较PAN组降低,差异具有统计学意义(P<0.05或P<0.01)。结论 BMP-7减轻尿液红细胞及尿蛋白的排泄,降低血清CysC、MIC水平,下调肾小球Wnt1/β-catenin信号通路的表达,减轻肾损伤。 Objective To investigate the effects of bone morphogenetic protein-7(BMP-7) on the expressions of glomerular Wnt1/β-catenin in rats with puromycin aminonucleoside-induced nephrosis(PAN).Methods Totally 20 rats with nephropathy induced by puromycin aminonucleoside were randomly divided into PAN group(PAN,n=10) and PAN +BMP-7 treament group(BMP-7,n=10).Rats in BMP-7 group were treated with intraperitoneal human recombinant BMP-7 injections of 30μg/kg twice a week.Another 10 normal rats were served as the control group(NC).Urinary erythrocytes(UE),24h urinay protein(24h UP),serum cystatin C(Cys C),macrophage migration inhibitory factor(MIF) were detected at 12 weeks.Rats were sacrificed to determine glomerular Wnt1 and β-catenin mRNA and protein expressions by RT-PCR and western blot,respectively.Results Compared with the NC group,the levels of UE,24h UP,Cys C,MIF,the mRNA and protein expressions of glomerular Wnt1 and β-catenin were significantly increased in the PAN group.The levels of UE,24h UP,Cys C,MIF,Glomerular Wnt1 and β-catenin mRNA and protein expression were decreased in the BMP-6 group compared with the PAN group.Conclusion BMP-7 can ameliorate the excretion of urinary erythrocyte and protein,decrease the level of serum Cys C and MIF,and down-regulate the expressions of glomerular Wnt1/β-catenin to alleviate renal injury.
