支架荷载的骨髓间充质干细胞的分化实验研究

摘要 目的探讨大鼠骨髓间充质干细胞(BMSCs)在改性聚乳酸-Ⅰ型胶原复合支架上向表皮干细胞分化的可行性。方法体外诱导改性聚乳酸-Ⅰ型胶原复合支架荷载的大鼠骨髓间充质干细胞向表皮干细胞分化,免疫组织化学法检测表皮干细胞标志物CK19和整合素β1的表达。结果诱导液作用2周后,免疫组织化学法检测到改性聚乳酸-Ⅰ型胶原复合支架荷载的大鼠BMSCs中大量阳性双表达CK19和整合素β1。结论改性聚乳酸-Ⅰ型胶原复合支架荷载的大鼠BMSCs仍具有向表皮干细胞分化的潜能,此支架具备作为组织工程皮肤支架材料的条件。 Objective To investigate the feasibility of Rat bone marrow mesenchymal stem cells(BMSCs) differeniated into epidermal cells on modified poly L acid and type collagen Ⅰ composites scafold(PLA-Ⅰ).Methods Differentiation of rat BMSCs on PLA-Ⅰ into epidermal stem cells in vitro by EGF induced cell culture solution.Two-signed of immunohistochemistry identifed CK19 and intergrin β1 markers epidermal stem cells.Results After two weeks of EGF induced cell culture solution,expression of CK19 and intergrin β1 was highly at rat BMSCs on PLA-Ⅰ.Conclusion Rat BMSCs still have the potential to differentiate into epidermal cells on PLA-Ⅰ,which can be used as a vehicle for tissue engineering skin.
