ω-3多不饱和脂肪酸对严重烧伤大鼠肠道炎症反应的影响 被引量:13

摘要 目的观察ω-3多不饱和脂肪酸对严重烧伤大鼠肠道炎症反应的影响,初步探讨其对肠道功能调节的机制。方法清洁级SD大鼠72只,随机分为3组:正常组(A组)、力能对照组(B组)和鱼油治疗组(C组)。复制大鼠30%TBSAⅢ度烧伤模型,A组大鼠经尾静脉注射生理盐水2 ml/(kg.d),B组每天定时经尾静脉注射20%力能中长链脂肪乳2 ml/(kg.d),C组在相同时间点经尾静脉注射与B组等氮等热量的尤文ω-3鱼油脂肪乳2 ml/(kg.d)。分别观察伤前(A组)、伤后(B、C组)1、4、7、10 d血清二胺氧化酶(DAO)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)水平;免疫组织化学染色法观察肠组织TNF-α的表达。结果B、C两组在伤后各时相点血清DAO、TNF-α、IL-6水平显著高于伤前(P<0.01);C组血清DAO、TNF-α、IL-6水平较B组降低,在伤后4、7、10 d与B组比较差异有显著性(P<0.05或P<0.01)。与B组相比,C组TNF-α活性明显减弱。结论ω-3鱼油脂肪乳降低烧伤后血清DAO水平,改善烧伤后肠黏膜损伤,保护肠黏膜屏障功能。ω-3鱼油脂肪乳减轻烧伤后促炎症介质TNF-α、IL-6的释放,抑制烧伤后全身炎症反应的程度。 Objective To investigate the effects of ω-3 polyunsaturated fatty acids(ω-3 PUFAs) on intestinal inflammatory responses following severely burned rats,and preliminary explore the adjustment mechanism of ω-3 PUFAs on the intestinal function.Methods A total of 72 SD rats with 30% total body surface area(TBSA) fully thickness burned were randomly divided into 3 groups: the normal control group(A group),the experimental group(B group) and the treatment group(C group).Rats of group A were injected NS with 2 ml/(kg·d) by tail vein,Rats of group B were injected 20% middle-long-chain fatty emulsion with 2 ml/(kg·d) by tail vein,Rats of group C were injected ω-3 fish oil with 2 ml/(kg·d) by tail vein at the same time for each day.The levels of serum diamine oxidase(DAO),tumor necrosis factor-alpha(TNF-α),inter leukins-6(IL-6) were measured before injury group(Group A) and after injury groups(Group B,C) in 1,4,7,10 d respectively.Expression of TNF-α in intestinal tissue was detected by immunohistochemical staining.Results Serum levels of DAO,TNF-α,IL-6 were significantly higher than those pre-injury at each time point after injury in group B and C(P0.01).The levels of DAO,TNF-α,IL-6 in group C were lower than those in group B,and were significantly different in 4,7,10 d after injury(P0.05 or P0.01).Compared with the group B,TNF-α activity was significantly decreased in group C.Conclusion ω-3 PUFAs can reduce serum DAO level,improve intestinal mucosa injury,and protect the function of intestinal mucosal barrier after burn.ω-3 PUFAs can reduce the releasing of pro-inflammatory media,such as TNF-α and IL-6,inhibit the extent of the systemic inflammation.
