COX-2、RAR-β_2在食管鳞状细胞癌组织中的表达及相互关系 被引量:11

Differential expression of COX-2 and RAR-β2 and their association in esophageal cancer tissue specimens

摘要 目的研究环氧化酶-2(COX-2)、维甲酸受体β2(RAR-β2)在食管鳞状细胞癌(ESCC)及周围正常黏膜组织中差异表达状况,探讨两者间相互关系及在ESCC中的作用。方法常规抽提33例食管癌及相应正常食管黏膜组织中总RNA,采用实时荧光定量PCR检测33例新鲜食管癌组织和正常黏膜组织中COX-2和RAR-β2基因的表达情况。结果与正常食管黏膜组织相比,食管癌组织中COX-2的表达明显上调,而RAR-β2的表达下调(P<0.05)。Ⅰ-Ⅱ级ESCC组织中RAR-β2mRNA的表达明显高于Ⅲ级ESCC组织(P<0.05);且RAR-β2mRNA的表达与患者是否喜好热烫饮食(T≥70℃)关系密切(P<0.05)。而COX-2 mRNA的表达与患者是否喜好腌制食物显著相关(P<0.05)。结论食管鳞状细胞癌与RAR-β2、COX-2的表达有密切相关性。 Objective To investigate the differential expression of COX-2,RAR-β2 in malignant and normal esophageal tissues,elucidate its correlation to some clinicopathologic parameters of esophageal squamous cell carcinoma and evaluate its clinic significance.Methods The expression of COX-2,RAR-β2 in 33 specimens of ESCC and corresponding noncancerous tissues were detected by real-time fluorescent quantitative polymerase chain reaction,and the relationship between COX-2,RAR-β2 and pathology of ESCC was further demonstrated.Results The expression of RAR-β2 of ESCC was significantly lower than that in normal tissues(P 0.05),and was significantly related to the tumor differentiation between the well-moderately and the poorly differentiation tissues(P 0.05),and was also related to the hot-drinking(T≥70℃)(P 0.05).The expression of COX-2 in ESCC was significantly higher than that in normal tissues(P 0.05),and was related to the pickled-food(P 0.05).They were not related with smoking excessive drinking,and the age and gender of the patients.Conclusion The ESCC is closely related to the expression of COX-2 and RAR-β2.
