血站利用O型血液中去除的淋巴细胞制备细胞因子诱导的杀伤细胞的可行性 被引量:3

摘要 目的探讨血站利用0型血液中去除的淋巴细胞制备细胞因子诱导的杀伤细胞(CIK)的可行性。方法采集30例0型献血者全血,利用白膜法4—6h内提取白细胞,然后用密度梯度离心法分离出淋巴细胞,再经分化、诱导、培养从而制备CIK细胞。结果30例0型献血者血液中去除的淋巴细胞制备CIK含量为(1.304-0.12)X1010/L,且cD3+/CD56+细胞表达占40%一60%,冰冻保存1个月后回收率达90%以上。结论利用0型无偿献血者血液中去除的淋巴细胞制备CIK,既利于保证原料血液的质量,又利于保证CIK细胞制品的质量和数量,可最大限度降低CIK的制备成本。 Objective To investigate the feasibility of preparing cytokine-induced killer cells (CIK) using the lymphocytes removed from O type blood in the blood bank. Methods The whole blood was collected from 30 blood donors with 0 type blood. The white blood cells were enriched and were seperated by density-grediant centrifugation. The lymphocytes were differentiated, induced and cultured to prepare CIKs. Results The CIKs produced from the 30 cases of blood donors with type O blood was ( 1.30 _+ 0. 12) x 1010+/L with CD3 +/CD56 + population up to 40% - 60%. The recovery rate was up to over 90% after 1 month of cryopreservation. Condusion Preparing the CIK with the lymphocytes removed from the type 0 blood donation is an ecnomical way to cut the cost of CIK preparation.
