摘要 目的探讨错配修复基因hMLH1在雌激素诱导结肠癌细胞凋亡中的作用。方法将含野生型hMLH1-cDNA的质粒转入hMLH1缺陷的人结肠癌细胞株HCT116,在不同浓度的雌激素干预下,分别以流式细胞仪和活细胞计数试剂盒检测转染后细胞的凋亡率和活性。结果与空载组相比,转染hMLH1后雌激素能诱导HCT116细胞活性为0.34±0.06,显著低于空载组的0.77±0.17(P<0.001),而凋亡率由9.75±0.53增加至45.20±2.63(P<0.001)。结论 hMLH1参与雌激素诱导的结肠癌细胞株HCT116凋亡。 Objective Exploring the role of mismatch repair gene hMLH1 played in the apoptosis of estrogen-induced colon cancer cell HCT116.Methods Transmit wild-type hMLH1-cDNA plasmid into hMLH1-deficient colon cancer cells(HCT116 cells) and then treat cells with different concentrations of estrogen.Detect the rate of apoptosis and the activity of cells with flow cytometry and cell counting kit-8 respectively.Results After transfection of hMLH1,estrogen induces the activity of HCT116 cells as 0.34 ± 0.06 that were significantly lower than those in no-load group 0.77 ± 0.17(P0.001).However,the apoptosis rate increased from 9.75 ± 0.53 to 45.20 ± 2.63(P0.001).Conclusions hMLH1 is involved in estrogen-induced apoptosis of colon cancer cell line HCT116.
