泻心汤有效成分对高糖诱导的肾小球系膜细胞的保护作用 被引量:1

摘要 目的研究泻心汤有效成分对高糖诱导的肾小球系膜细胞模型的影响。方法体外高糖(30 mmol·L^(-1))培养肾小球系膜细胞,分别加入不同浓度的小檗碱、黄芩苷和大黄提取物,孵育24 h后收集细胞上清液,采用MTT法测定细胞相对活力,ELISA测定Ⅵ型胶原及单核细胞趋化蛋白1(MCP-1)的浓度,利用比色法检测丙二醛(MDA)的含量及超氧化物歧化酶(SOD)活力。结果小檗碱、黄芩苷和大黄提取物均可剂量依赖性抑制高糖诱导的肾小球系膜细胞增殖、Ⅳ型胶原及MCP-1的分泌,提高SOD的酶活力和减少MDA的产生。结论在高糖诱导的肾小球系膜细胞中,泻心汤有效成分具有独立于降糖调脂的直接肾小球系膜保护作用。 AIM To investigate the effects of ingredients of Xiexin decoction on glomerular mesangial cells under high glucose condition. METHODS Rat mesangial cells(HBZY-1)were cultured and treated with 30 mmol· L^- 1 glucose and different concentrations of berberine, baicalin or rhubarb extracts (containing 35.7 % polysaccharides), respectively for 24 h. Cell preliferation was measured by MTr assay. Collagen IV and monocyte chemotactic protein (MCP-1) were examined by ELISA. MDA as well as SOD activities were measured by spectmphotometry. RESULTS Berberine, baicalin and rhubarb extracts could inhibit cell proliferation, collagen iV and MCP-1 production, reduce MDA level and elevate SOD activities, respectively in a dose-dependent manner in rat mesangial cells under high-glucose condition. CONCLUSION The ingredients of Xiexin decoction could directly protect glomemlar mesangial cells which is independent from their hypoglycaemic and lipid-lowering effects.
