亚低温对急性脊髓损伤后肿瘤坏死因子-α表达及运动功能恢复的影响 被引量:6

摘要 目的观察大鼠脊髓损伤(SCI)后亚低温对肿瘤坏死因子-α(TNF-αmRNA表达及运动功能恢复的影响,探讨其可能的作用机制。方法72只SD大鼠随机分为对照组(/Z=24)、常温组(n=24)和亚低温组(n=24),每组再分为六个亚组,每亚组4只,参照改良Alien法建立大鼠脊髓(19)中度损伤模型,亚低温组给予亚低温治疗5h,而对照组不做亚低温处理。分别于损伤后6h、12h、24h、72h、1周和4周,利用Tarlov评分检测亚低温对大鼠SCI后运动功能恢复的影响;然后将大鼠处死,利用半定量RT-PCR方法观察损伤段脊髓组织中TNF-amRNA表达的变化。结果亚低温组TNF-dmRNA表达SCI后6-72h明显少于常温组(P〈0.05),而运动功能评分每个时间点都明显高于常温组(P〈0.05)。结论亚低温可明显抑制SCI后TNF-α的表达,具有良好的恢复运动功能的作用。 To investigate the effectiveness of mild hypothermia therapy on the expression of tumor necrosis factor -α (TNF -α) and neurological outcome after spinal cord injury (SCI) in rats and to discuss the possible mechanism. Methods 72 SD rats were divided randomly into the normal control group( n = 24 ) , normal temperature group (n = 24 ) and mild hypothermia group( n = 24). Each group was subdivided into 6 subgroups according to the post SCI time, with 6 SD rats in each sub group. The moderate SC1 models were made with SD rats according to modified Allen "s method. The rats in mild hypothermia group were cooled by mild hypothermia for 5 hours, while those in other two groups were subjected to no disposal. The neurological outcome of rats in every subgroup was evaluated with Tarlov scores, then the rats was killed at the points of 6 h,12 h, 24 h, 72 h, 1 w and 4 w after SCI, respectively. Semiquantitative RT PCR method was performed to confirm the mRNA expressions of TNF - ct in the spinal cord of rats. Results The expression of the TNF -α mRNA and the neurological score in rats of the mild hypothermia group was less than that of the SCI group at each time point, there was significant difference in the expression of the TNF-α mRNA among the 12 h, 24 h and 72 h subgroups(P 〈0. 05) ; and there was also significant difference in the neurological score in every time subgroups (P 〈 0. 05). Conclusion Hypothermia can inhibit the expression of the TNF-α and improve motor function obviously.
