摘要
目的获取正常C57BL6小鼠的呼吸系统组织学图谱资料,并与人体呼吸系统的组织学特征进行比较。方法选取从南京大学模式动物研究所购得的C57BL/6J小鼠20只,在SPF级环境中用全价营养鼠饲料饲养,饲养到6周龄时将其处死。在1 h内对处死的小鼠进行组织解剖,取出呼吸系统器官和组织,于10%中性缓冲福尔马林固定;常规病理制片,HE染色;显微镜观察,照相机摄影,图像采集,用计算机对图像进行标注。结果对小鼠呼吸系统进行详细、系统的组织学观察,并发现人体与小鼠在呼吸系统的差异主要存在于气管腺,小鼠气管腺分3种:浆液腺、粘液腺及混合腺,主要分布于粘膜固有层;人的气管腺主要为混合腺,分布于粘膜下层。结论正常C57BL6小鼠的呼吸系统组织学图谱已基本建立,为利用该品系小鼠建立呼吸系统疾病遗传工程小鼠模型奠定了基础。
Objective To establish of a histology atlas for wild type C57BL6 mouse respiratory system and to compare histological char acteristics of respiratory system between human and mice. Methods Totally 20 Wild-type C57BL6 mice were purchased from model animal research centre in Nanjing university and housed under specific pathogen-free conditions. Adult mice of six weeks old were killed and tissue samples of respiratory system were collected and fixed into 10% PBS-buffered formalin. The fixed samples were then subjected to conventional sections and HE staining. Histological figures were taken using Olypmus microscope equipped with Olympus D300 camera and marked using photoshop software. Results Detailed and systematic histological investigation was performed using mouse respiratory tissues. The main histological difference between the respiratory systems of human and mice existed in tracheal glands. Mice have tracheal glands of three different types : serous gland, mucous gland and mixed gland, which are located in mucous membrane proper. In contrast, human tracheal glands are mainly of one type---mixed gland,which is mainly located in submucous layer. Conclusion The histological atlas of mouse respiratory system is constructed, which can facilitate the generation of novel genetically manipulated mouse disease models of respiratory system.
