摘要
目的初步探寻红葡萄酒中主要活性成分白藜芦醇对紫外线照射皮肤保护作用的可能机制。方法白藜芦醇对UVB照射的人皮肤成纤维细胞基质金属蛋白酶(MMP)-1水平及细胞凋亡的影响。将培养的人皮肤成纤维细胞分为5组,A组:无UVB照射无干预组,B组:UVB照射无干预组,C组:UVB照射1μmol/L白藜芦醇干预组,D组:UVB照射10μmol/L白藜芦醇干预组,E组:UVB照射100μmol/L白藜芦醇干预组。UVB照射剂量均为30mJ/cm2。用免疫组织化学方法分别检测5组细胞中MMP-1水平,用磷脂酰丝氨酸外翻分析(AnnexinV法)检测细胞凋亡率。结果 A~E组5组细胞MMP-1阳性细胞评分及凋亡率差异均有统计学意义(P均<0.05)。A组评分及凋亡率均明显低于B组,两组比较差异有统计学意义(P均<0.05),B组与C,D,E组评分及凋亡率比较差异均有统计学意义(P均<0.05),C~E组3组评分及凋亡率比较差异亦有统计学意义(P<0.05)。结论白藜芦醇可以抑制UVB照射诱导的人皮肤成纤维细胞MMP-1水平的升高,并可减少细胞因UVB照射引起的凋亡,白藜芦醇可能通过此机制对紫外线照射皮肤起到保护作用。
Objective In order to explore the protective effect of resveratrol,the main active ingredient in red wine,to skin from UVB radiation. Methods The level of MMP-1 and apoptosis of human skin fibroblasts before and after UVB exposure were compared. Human skin fibroblasts were divided into 5 groups. Group A was no UVB radiation and no intervence group,group B was irradiated by UVB (30mJ/cm2) without intervence, group C-E were irradiated by UVB (30mJ/em2) and were interposed by 1μmol/L, 10μmol/L and 100μmol/L resveratrol respectively. The MMP-1 levels of the 5 groups were detected by immunohistochemistry. The cell apoptosis rates of the 5 groups were detected by Annexin V method. Results There were significant difference of the MMP-1 positive cells score and apoptosis rate among the 5 groups (P 〈 0.05 respectively). The MMP-1 positive ceils score and apoptosis rate in group A were lower than those in group B (P 〈 0.05). There were significant difference of the MMP-1 positive cells score and apoptosis rate among the B-E groups (P〈 0.05 respectively). And the MMP-1 positive cells score and apoptosis rate of C-E groups had significant difference (P 〈 0.05 respectively). Conclusion The increase of MMP-1 in human skin fibroblasts after UVB irradiation can be restrained by resveratrol. Also resveratrol can reduce the cell apoptosis caused by UVB irradiation. It maybe the mechanism of protective affects that resveratrol act in human skin radiated by ultraviolet rays.
