摘要
目的:建立新的测定鬼臼毒素纳米脂质载体包封率的高效液相色谱法。方法:以ODS-C18(4.6 mm×250 mm5,μm)为分析柱,以甲醇-0.6%冰醋酸(52∶48)为流动相,流速1 mL.min-1,检测紫外线波长为291 nm,进样量20μL,柱温40℃。采用4℃12 000 r/min离心30分钟分离脂质体和游离药物,测定包封率。结果:当鬼臼毒素浓度在0.5~200μg/mL范围内时,峰面积与浓度之间呈良好的线性关系(r=0.999 1,n=8),高、中、低3个剂量的回收率分别为98.6%,96.98%,98.6%,日内精密度0.735%3,批鬼臼毒素纳米脂质载体中鬼臼毒素的包封率分别为84.9%、80.9%8、3.0%。结论:高速离心-HPLC法适合测定0.5%鬼臼毒素纳米脂质载体中鬼臼毒素的包封率。
Objective:To establish an HPLC method for the determination of entrapment efficiency of Podophyllotoxin-Nanostructured lipid carriers(POD-NLC). Methods:An analytic column of ODS-C18 column(4.6 mm×250 mm,5 μm) was used with methanol-0.6% glacial acetic acid(52∶48) as the mobile phase at a flow rate of 1.0 mL/min.The UV detection was set at 291 nm.The column temperature was 40℃.Entrapment efficiency was determined after separation of the unentrapped drug by centrifugation at 12 000 r/min for 30 min at 4℃. Results:A good linear relationship was found between peak area and the concentration of POD in the range of 0.5~200 μg/mL(r=0.999 1,n=8).The sample recovery rates were 98.6%,96.98%,98.6%,respectively for high,moderate and low doses,with RSD of 0.735%(n=3).The entrapment efficiency of 3 paths of POD-NLC were 84.9%、80.9%、83.0% respectively. Conclusion:It was suitable to assay the entrapment efficiency of 0.5% POD-NLC with the high-speed centrifugation-HPLC method.
