摘要
用HPLC法测定肉苁蓉中麦角甾苷的含量.采用Kromasil C18(4.6 mm×250 mm,5μm)色谱柱,以甲醇-水(V(甲醇)∶V(水)=35∶65)为流动相(水中含有0.4 mL/L磷酸)等质洗脱,流速1 mL/min,检测波长331 nm,柱温35℃.检测结果表明:麦角甾苷在0.34~2.04μg范围内呈良好线性关系,回归方程为A=192.57C-2.896(r=0.999 9),平均回收率为98.1%,RSD为1.99%(n=5).本方法用于测定肉苁蓉中麦角甾苷的含量,重现性良好、结果准确可靠.
A HPLC method for simlultaneous determination of acteoside in herbe cistanches was established.Agilgent 1100 system and Kromasil C18(4.6 mm×250 mm,5 μm)were adopted.The mobile phase consisted of H2OCH3OH(65∶35)and the detection wavelenth was 331 nm with a PDA detector.The flow rate was 1 mL/min and the column temperature was 35 ℃.The linear ranges of acteoside was 0.34~2.04 μg,the average recovery was 98.1 %,RSD was 1.99 %(n=5).This method is accurate and reliable.It can be used to determine the content of acteoside in herbe cistanches.
