酪氨酸蛋白激酶研究中磷酰酪氨酸的快速分离与鉴定

A RAPID AND SIMPLE METHOD FOR SEPARATION OF PHOSPHOTYROSINE FROM PHOSPHOSERINE AND PHOSPHOTHREONINE

酪氨酸蛋白激酶是与生长调控有关的重要酶类。分离磷酰酪氨酸与其它磷酰氨基酸(磷酰丝氨酸、磷酰苏氨酸)是测定酪氨酸蛋白激酶的关键之一。本研究用醋酸纤维素薄膜电泳法与放射自显影法相结合以快速分离、并鉴定磷酰酪氨酸,获得了理想的效果。

The seperation of phosphotyrosine from phosphoserine and phosphothreonine is essential for determination of tyrosine protein kinase activity, one of the important enzymes which are closely related to cell growth regulation system.In this study, by electrophoresis on acetate cellulose membrane and autoradiography technique, the spot of phosphotyrosine was showed easily separated from those of pnosphoserine, phosphoth-treonine and free phosphate.
