摘要
目的探索脂质体、CpG寡核苷酸(ODN)以及Hepa 1-6肝癌细胞裂解物联合应用治疗肝癌的免疫效果。方法建立C57BL/6小鼠肝癌模型,以PBS、ODN+Hep(肝癌细胞Hep)、Lip(Lipofectamine脂质体)+HDN+Hep荷瘤鼠背部皮下注射,各组小鼠肝脏HE染色,FCM检测小鼠脾脏CD3-FITC、CD4-FITC、CD8-FITC、NK1.1-FITC的组成,ELISA法测免疫小鼠血清中IFN-γ、IL-4水平。结果脂质体包裹CpG ODN+Hepa 1-6肝癌细胞裂解物中CD3-FITC、CD4-FITC、CD8-FITC、NK1.1-FITC的体外杀伤活性以及荷瘤小鼠血清中IFN-γ、IL-4水平均显著高于CpG ODN+Hep组以及PBS组。结论脂质体包裹CpG ODN+Hep能显著提高机体的免疫功能,尤其是特异性细胞免疫功能。
Objectives This article discussed the immune effect of combination application of liposomal, CpG ohgodeoxynacleotide ( ODN ) and Hepa I - 6 hepatoma cell lysates for liver cancer treatment. Methods Liver cancer model of C57BL/6 mice was set up. Tumor-bearing mice were sub- cutaneously injected PBS, ODN + Hep, Lip + ODN + Hep treatment and observation. Liver of different group mice used HE staining. FCM detect- ed the formation of spleen CD3 - FITC, CIM - FITC, CD8 - FITC, NK1.1 - FITC. The serum of mice immunized with IFN - y, I L - 4 level was measured by ELISA method. Results The destruction to competence in vitro of liposomes coating CpG ODN + Hepa 1 - 6 and CD3 - F1TC,CD4 - FITC , CD8 - FITC, NK1.1 - FITC in lysate of liver cancer cells and level of IFN - y, IL - 4 in mice serum are much higher than in CpG ODN + Hep group and PBS group. Conclusions Liposomal CpG ODN and Hep can significantly improve immune function, especially the specific cellular immune function.
