NGF基因转染对人牙周膜细胞生物学特性的影响

摘要 目的探讨神经生长因子(NGF)基因转染对牙周膜细胞(PDLCs)生物学特性的影响。方法体外培养PDLCs,分为NGF转染组、转染空载体组、对照组。运用基因转染技术将NGF基因通过质粒pcDNA3.1-NGF转染入PDLCs中,采用免疫组化及免疫印记法鉴定转染细胞的表达,MTT法观察其增殖能力、碱性磷酸酶(ALP)活性。结果免疫组化证实,NGF基因转染PDLCs中有NGF基因表达。与转染空载体组和对照组比较,NGF转染组可见PDLCs明显增殖,转染NGF基因后PDLCs的ALP活性较另两组明显增高(P均<0.05)。结论 NGF基因转染的PDLCs有成骨细胞活性,并能使细胞增殖为牙周组织再生的种子细胞。
