ω-3PUFA对颅脑损伤伴休克大鼠早期肺损伤的保护作用 被引量:2

摘要 目的观察ω-3多不饱和脂肪酸(ω-3 PUFA)对颅脑损伤伴失血性休克(TBIS)大鼠早期肺损伤的保护作用。方法 Wistar大鼠36只,随机分成假手术组(C组)、TBIS模型组和ω-3 PUFA治疗组,每组12只。检测血清肿瘤坏死因子(TNF)-α和肺组织超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽(GSH)的含量;计算肺湿干重比(W/D);光镜下观察肺组织病理变化,并做病理评分。结果与C组比较,TBIS组与ω-3 PUFA治疗组肺组织匀浆MDA及血清TNF-α水平、肺组织W/D及损伤评分均明显升高(P<0.01);肺组织匀浆SOD及GSH活性均降低(P<0.01)。与TBIS组比较,ω-3 PUFA治疗组血清TNF-α及肺组织匀浆MDA水平、肺组织W/D及损伤评分均明显降低(P<0.01或P<0.05),SOD及GSH活性均升高(P<0.01)。结论ω-3 PUFA预处理可减轻TBIS大鼠早期肺损伤,可能与其抑制氧自由基产生及促炎细胞因子的释放有关。 Objective To investigate the protective effects of ω-3 polyunsaturated fatty acid(ω-3 PUFA) on early pulmonary injury in rats with traumatic brain injury and hemorrhagic shock(TBIS).Methods A total of 36 male Wistar rats were assigned randomly into 3 groups(12 in each): sham operation group(group C),TBIS model group and PUFA pretreatment group.The arterial blood samples were taken for determination of serum levels of tumor necrosis factor-α(TNF-α).The pulmonary tissues were taken for the determination of levels of superoxide dismutase(SOD),malondialdehyde(MDA) and glutathione(GSH),and microscopic examination.Results Compared with group C,the serum levels of TNF-α,MDA level in pulmonary tissues,W/D lung weight ratio and pathologic scores of pulmonary tissues were significantly higher(P0.01),and SOD and GSH in group TBIS and group PUFA were significantly lower(P0.01).The serum level of TNF-α,MDA level in pulmonary tissues,lung W/D weight ratio and pathologic scores of lung tissue were significantly lower(P0.01 or P0.05),and the levels of SOD and GSH were significantly higher(P0.01) in group PUFA than those in group TBIS.Conclusion The supplementation of ω-3 PUFA can ameliorate early pulmonary injury in rats with TBIS,which may contribute to the inhibition of expression of oxygen free radicals and inflammatory cytokines.
