龙葵糖蛋白对MCF-7细胞内[Ca^(2+)]_i的影响 被引量:3

摘要 研究龙葵糖蛋白对乳腺癌MCF-7细胞内[Ca2+]i的影响.通过MTT法测定龙葵糖蛋白对MCF-7细胞的细胞毒作用;Hoechst33258染色荧光显微镜下观察龙葵糖蛋白作用MCF-7细胞的形态学变化;采用Fluo-3/AM探针标记,激光共聚焦技术观测龙葵糖蛋白对MCF-7细胞内[Ca2+]i的影响.结果表明,龙葵糖蛋白对MCF-7细胞有抑制作用,其IC50为973.46μg/mL;Hoechst33258荧光染色结果显示,龙葵糖蛋白与MCF-7细胞共培养后,随着剂量增加细胞呈明显的固缩状,细胞核出现碎片和凋亡小体等细胞凋亡现象;激光共聚焦检测显示龙葵糖蛋白可升高MCF-7细胞内钙离子的质量浓度,并且随着龙葵糖蛋白剂量的增加,细胞内钙离子质量浓度也相应地增大,且与对照组相比均具有统计学意义(P<0.01):龙葵糖蛋白能够升高人乳腺癌MCF-7细胞内[Ca2+]i的质量浓度. To study the effects of glycoprotein isolated from solanum nigrum L.(SNL glyco-protein) to human breast cancer MCF-7 cells [Ca2 +]i.MTT was adopted to determine ifSNL glycoprotein had cytotoxic effect to MCF-7;Hoechst33258 staining was observed underfluorescence microscope role of SNL glycoprotein MCF-7 cell morphology;by Fluo-3/AMprobe labeling,confocal observation effects of SNL glycoprotein on MCF-7 cells [Ca2+]i.The results showed that the SNL glycoprotein on the inhibition of MCF-7 cells,the IC50 was973.46 μg/mL;Hoechst33258 staining showed that the obvious appearance of apoptosis:nuclear condensation and fragmentation,chromatin condensation,and apoptotic body forma-tion;laser confocal analysis showed that SNL glycoprotein can increase MCF-7 intracellularcalciumion concentration,and with the dose of SNL glycoprotein increase in intracellular cal-cium concentration increases accordingly,compared with the control group were statisticallysignificant(P 0.01).SNL glycoprotein to increase breast cancer cell line MCF-7 cellscalcium concentration.
