蓝舌病毒湖北株对人前列腺癌PC-3细胞杀伤效应的研究

摘要 目的体外实验研究蓝舌病毒湖北株3(BTV-HbC3)对人前列腺癌PC-3细胞的杀伤效应并且初步探讨BTV-HbC3靶向性溶瘤的机制。方法观察PC-3细胞感染BTV-HbC3后所致的病变效应(CPE)变化;透射电镜观察细胞感染病毒后超微结构的变化;CCK-8法研究病毒致细胞病变效应的特征;DNA Ladder分析病毒诱导细胞凋亡的情况;流式细胞仪检测病毒对PC-3细胞凋亡的影响。结果 BTV-HbC3感染PC-3细胞后可见到明显的细胞效应;透射电镜发现胞浆内有大量病毒颗粒和典型细胞凋亡的形态变化;CCK-8法检测到病毒明显抑制细胞生长;DNA Ladder分析见阶梯状条带;流式细胞仪检测到明显细胞凋亡的存在,可见明显的亚二倍体峰。结论 BTV-HbC3在体外可有效地杀伤PC-3细胞,并能诱导其凋亡。 Objective To investigate the characteristics and the mechanism of bluetongue virus strain HbCS; (BTV-HbC:,) infccting PC-3 malignancy in vitro. Methods PC-3 cells were infected with BTV-HbC3 to observe the cytopathic effects (CPE). Changes of ultrastructurc of the infectedccllswercobscrvcd with transmission electron microscope (TEM). The inhi- bition activity of the infected PC-3 cells was analyzed with CCK-8. The apoptosis of PC-3; cells induced by BTV-HbC:, was de- tected with DNA ladder. The effect of virus on the apoptosis of PC-3 cells was detected by flow cytomctry (FCM). Results Obvious CPE was observed. TEM observation found lots of virus particles in the cytoplasm and typical changes of ultrastruc- ture. CCK-8 detected that the virus could significantly inhibit the growth of cells. DNA ladder found ladder strip shape. FCM detected significant cell apoptosis with hypodiploid peak. Conclusions BTV-HbC-3; is able to infect and replicate in PC-3 cells effectively in vitro and induce tumor cell apoptosis.
