奈达铂与顺铂对人肝肾细胞损伤作用的对比 被引量:8

摘要 目的对比奈达铂(NDP)与顺铂(DDP)对人肝细胞HL-7702、人胚肾细胞293T的不良反应。方法采用四甲基偶氮唑盐(MTT)法测定不同浓度的NDP和DDP对HL-7702、293T细胞的抑制作用。流式细胞仪(FCM)检测加NDP和DDP前后,HL-7702、293T细胞凋亡率的变化。测定HL-7702、293T细胞损伤前后细胞的活性氧自由基(ROS)及细胞裂解液中丙二醛(MDA)的含量及超氧化物歧化酶(SOD)的活性。结果 MTT显示不同浓度的NDP和DDP对HL-7702、293T细胞均有一定的抑制作用,且DDP对HL-7702、293T细胞的损伤作用明显高于NDP(P<0.05)。FCM也表明DDP对HL-7702、293T细胞的抑制作用较NDP明显。NDP和DDP均能导致HL-7702、293T细胞ROS、MDA含量增加,SOD活性降低。DDP对HL-7702、293T细胞的氧化损伤更明显(P<0.05)。结论 NDP和DDP对HL-7702、293T细胞均有细胞毒性和氧化损伤作用。NDP对HL-7702、293T细胞的损伤作用较DDP小。
