摘要
目的检测FoxO1在食管鳞状细胞癌组织中的表达水平,分析其与食管鳞状细胞癌各临床病理因素的关系。方法应用实时荧光定量PCR SYBR Green荧光染料法,检测42例手术切除的食管鳞癌组织及配对的正常食管黏膜组织中FoxO1 mRNA的表达;免疫组化S-P法检测FoxO1蛋白的表达及组织学定位。结果正常食管黏膜组织中FoxO1mRNA及蛋白均呈低表达,而食管鳞癌组织中FoxO1 mRNA及蛋白表达显著升高(P<0.01)。高分化食管鳞癌组织的相对表达量显著高于中低分化食管鳞癌组织(P<0.01)。食管鳞癌组织中FoxO1 mRNA及蛋白表达与患者性别、年龄、肿瘤发生部位、TNM分期及有无淋巴结转移无明显相关性(P>0.05)。结论 FoxO1在正常食管黏膜组织和食管鳞癌组织中的表达存在明显差异,其表达水平与肿瘤分化程度密切相关。
Objective To determine the expression of transcription factor FoxO1,and its correlations to the clinicopathological features of esophageal squamous cell carcinoma(ESCC).Methods SYBR green real-time fluorescent quantitative reverse transcriptase PCR was used to detect the expression levels of FoxO1 mRNA in 42 matched pairs of ESCC tissues and normal samples.The protein expression and location of FoxO1 were measured by immunohistochemistry(SP method).The expression was analyzed with gender,age,site,TNM stage,lymphatic metastasis,and tumor cell differentiation.Results The expression of FoxO1 at mRNA or protein levels was upregulated significantly in ESCC tissues compared with normal esophageal epithelium tissues(P0.01).The expression of FoxO1 was higher in well differentiated ESCC than in poorly differentiated ESCC and normal esophageal epithelium tissues(P0.01).However,the expression of FoxO1 was not correlated to clinicopathological characters,such as gender,age,tumor site,TNM stage,lymphatic metastases and tumor cell differentiation(P0.05).The protein expression of FoxO1 was consistent with that of its mRNA expression.Conclusion The expression of FoxO1 varies significantly in ESCC and normal esophageal epithelium tissues.Over-expression of FoxO1 is closely related to the differentiation of ESCC.
