摘要
目的研究针刺督脉经穴对局灶性脑缺血大鼠脑组织核转录因子κB(NF-κB)蛋白表达的影响。方法将雄性SD大鼠随机分为假手术组、模型组、针刺对照组和针刺督脉组共四组,线栓法制作大脑中动脉栓塞(MCAO)模型。利用免疫组化法检测针刺后NF-κB蛋白表达的变化;以激光共聚焦扫描显微镜检测NF-κB转录活性。结果大鼠MCAO后NF-κB转录和蛋白表达增加,针刺督脉组MCAO 24 h和72 h NF-κB蛋白表达减少,在MCAO 24 hNF-κB转录活性减弱。结论针刺督脉经穴可能通过抑制MCAO急性期NF-κB蛋白的过度表达,减少细胞凋亡,减轻炎症反应,改善脑缺血再灌注损伤。
Objective To investigate the effect of acupuncturing Governor Vessel on expression of NF-κB for ischemic cerebrovascular disease in rats.Methods Male SD rats were randomly divided into 4 groups.The MCAO rats were prepared by suture method.The animals in therapy group received acupuncturing Governor Vessel.The expression of NF-κB has been determined.Results The expression of NF-κB was significantly higher after MCAO.The expression of NF-κB at 24 h and 72 h after MCAO in therapy group was lower than that in MCAO group.The transcriptive activity of NF-κB at 24 h after MCAO in therapy group was weaker than that in MCAO group.Conclusion Acupuncturing Governor Vessel may reduce expression of NF-κB during acute phase of MCAO.It may be related to the mechanism of acupuncturing on Governor Vessel therapy to ischemic cerebrovascular disease.
