VEGF-siRNA在肾癌中的实验研究

摘要 目的研究血管内皮生长因VEGF在肾癌细胞中的表达,检测对肾肿瘤的影响。方法将针对血管内皮生长因子的RNA的模板区的siRNA转导肾癌细胞786-0中,采用免疫组化检测血管内皮因子VEGF的蛋白的表达,通过siRNA对血管内皮生长因子的抑制,减少肿瘤中血管的生成,从而抑制小鼠肿瘤的生长。结果通过siRNA对肾癌血管内的血管内皮生长因子的干扰,抑制了肿瘤血管的生长,减慢了肿瘤的生长速度。结论 siRNA抑制血管内皮生长因子的表达,能明显减慢肿瘤的生长速度;肿瘤的生长速度与VEGF过度表达有关。 Objective To investigate the expression of vascular endothelial growth factor(VEGF) in renal carcinoma cell,detect the influence to renal carcinoma.Methods siRNA contraposed RNA of VEGF in template area was transducted in 786-0 of renal carcinoma cell,detected albumen expression of VEGF adopt immunohistochemistry,reduced angiogenesis in tumour through inhibiting VEGF by siRNA,inhibited tumor growth of mouse.Results Through obstructing VEGF in artery of renal carcinoma by siRNA,tumor vessel growth had been inhibited,the velocity of tumor vessel growth had been revved down.Conclusion siRNA can inhibit the expression of VEGF,down the velocity of tumor vessel growth which has relation with overexpression of VEGF.
