摘要
目的:研究X射线对稳定转染鸟苷解离抑制因子(LyGDI)A549细胞株放射敏感性的影响及相关机制。方法:应用生长曲线观察细胞增殖能力,流式细胞术检测不同剂量照射后转染组和对照组细胞的周期分布及凋亡率,克隆形成试验观察转染组细胞放射敏感性。结果:转染组细胞相对于对照组增殖能力减弱,F=212.55,P<0.01;克隆形成能力减弱,放射敏感性增加,F=7.56,P<0.05;流式细胞术检测到辐射诱导凋亡增加,F=18.45,P<0.01;细胞周期检测提示转染组G2期阻滞受抑制,F=6.29,P<0.05。结论:过度表达的LyGDI上调A549细胞放射敏感性,其可能机制为抑制G2期阻滞促进凋亡。
OBJECTIVE:To investigate the effect of X-ray on radiosensetivity in A549 cell line of stable transfected with LyGDI and the relative mechanism.METHODS: Growth curve was used to measure changes of cell proliferation.Flow cytometry was used to determine cell cycle distribution and apoptotic cell rate.Clony formation assay was used to measure radiosensetivity of the A549 with different phenotypes.RESULTS: The cell line of stable transfected with LyGDI got a lower proliferation(F=212.55,P0.01) and survival rate(F=7.56,P0.05) after irradiation with different doses of X-ray compared with control cell line.Radiation-induced G2 arrest was abrogated in A549-LyGDI cell line(F=6.29,P0.05) and the apoptosis induced by X-irradiation in A549-LyGDI line was enhanced(F=18.45,P0.01).CONCLUSIONS: It showed that LyGDI induced cell apoptosis upgraded sensitivity to radiation in A549 cell line.Abrogation of radiation-induced G2 arrest can be part of the mechanism.
