摘要
目的:观察三化汤对大鼠脑缺血再灌注血脑屏障损伤的保护作用。方法:将大鼠随机分为假手术组、模型组、三化汤低、高剂量组(7.2,14.4 g.kg-1)ig、尼莫地平组(8.1 mg.kg-1)ig。大鼠常规饲养3 d后灌胃给药,每日1次,连续给药7d后采用线栓法制备脑缺血再灌注大鼠模型。采用比色法检测大鼠脑组织伊文思蓝(EB)的含量,采用酶联免疫法检测大鼠血清S100B蛋白的含量。结果:假手术组大鼠脑组织EB含量、血清S100B蛋白含量减少,模型组大鼠脑组织EB含量、血清S100B蛋白含量显著升高(P<0.01);与模型组比,各用药组大鼠脑组织EB含量、血清S100B蛋白含量显著降低(P<0.05),其中三化汤高剂量组、尼莫地平组比三化汤低剂量组降低明显(P<0.05)。结论:大鼠脑缺血再灌注后可引起血脑屏障的损伤,三化汤对大鼠脑缺血再灌注后血脑屏障损伤具有一定的保护作用。
Objective:To observe the protective effect of Sanhua Tang on blood brain barrier injury in cerebral ischemia-reperfusion rat.Method:The rats were randomly dividec into sham operation group,model group,low dose group of Sanhua Tang,high dose group of Sanhua Tang and nimodipine group.Blocking middle cerebral artery of rat with suture method was used to prepare cerebral ischemia reperfusion model.The content of EB in rat brain tissue was detected by colorimetry.The content of S100B protein was detected in serum of cerebral ischemia-reperfusion rat by enzyme linked immunoassay.Result: In sham operation group the content of EB,S100B protein was decreased.The content of EB,S100B protein in model operation group was significantly increased(P0.01);compared with model operation group,in drug group the content of EB,S100B protein was significantly decreased(P0.05);in large dose group of Sanhua Tang and nimodipine group the content of EB,S100B protein was more significantly reduced than other groups(P0.05).Conclusion: Sanhua Tang can protect blood brain barrier injury of cerebral ischemia reperfusion rat.
