复方扶芳藤合剂对小鼠外周血干细胞动员作用的研究

The Study of Periphearal Blood Stem Cell Mobilization by Compound Fufangteng Mixture in Mice

目的:运用复方扶芳藤合剂对小鼠进行干预,观察复方扶芳藤合剂对小鼠外周血干细胞数量的影响,证明复方扶芳藤合剂对小鼠外周血干细胞具有从骨髓到外周血的动员作用。方法:取昆明种小鼠随机分为4组,分别为生理盐水组(空白组)皮下注射生理盐水15 mL.kg-1,同时生理盐水25 mL.kg-1ig;复方扶芳藤合剂组(药物干预组)复方扶芳藤合剂15mL.kg-1ig,同时sc生理盐水25 mL.kg-1;重组粒细胞集落刺激因子(G-CSF)组(阳性对照组)sc G-CSF 10μg.kg-1,同时生理盐水25 mL.kg-1ig;G-CSF加复方扶芳藤合剂组(联合用药组)复方扶芳藤合剂15 mL.kg-1.d-1ig,同时sc G-CSF 10μg.kg-1。给药6 d后,取静脉血检测外周血白细胞、单个核细胞数量,用甲基纤维素微量培养法培养进行混合细胞集落形成单位(CFU-Mix)集落计数,流式细胞术检测外周血干细胞抗原-1(Sca-1+)细胞数量和百分率。结果:药物干预组、阳性对照组和联合用药组的小鼠外周血白细胞数量明显高于空白组。药物干预组、阳性对照组和联合用药组经体外造血祖细胞培养CFU-Mix产率显著高于空白组,CFU-Mix分别为9,16,19个(P<0.05)。同时药物干预组、阳性对照组和联合用药组外周血中Sca-1+细胞百分率比空白组明显提高,分别为0.79%,1.15%,1.72%(P<0.05)。结论:复方扶芳藤合剂对小鼠外周血造血干细胞有明显的动员作用,具有作为有效的干细胞动员剂的潜力;复方扶芳藤合剂和G-CSF联合动员的效果好于单用复方扶芳藤合剂或单用G-CSF的动员效果,这提示在今后的应用中可考虑G-CSF联合复方中药进行外周血干细胞的动员。

Objective：To study the mobilization of peripheral blood stem cell in mice by compound fufangteng mixture.Method： Sixty mice were divided into four groups at random（n=15 each）.Control group was treated with normal saline;fufangteng mixture group was treated with fufangteng mixture,granulocyte colony-stimulating factor（G-CSF） group was treated with G-SCF,and Fufangteng mixture combined G-CSF group group was treated withy G-SCF and Fufangteng mixture.The number of peripheral white blood cell（WBC） and mononuclear cell（MNC） was counted after 6 days,the number of colony-unit-mix（CFU-Mix） colony was cultured by methylcellulose micro-culture,and number and percentage of stem cell antigen 1（Sca-1） cells were counted by flow cytometry.Result： The number of peripheral WBC and MNC was higher than pre-treatment,exceptfor control group.The number of WBC was（7.5±0.9）,（9.0±0.6）,（16.1±1.4）,（19.8±0.4）×106/L（P0.05） respectively,while the number of MNC was（4.0±0.7）,（6.6±0.3）,（10.5±0.9）,（12.6±1.4）×106/L（P0.05） respectively.The yields of CFU-Mix were higher than control group.Meanwhile,the percentage of Sca-1＋ in peripheral blood stem cells was higher than control group,and the percentage was 0.79%,1.15%,1.72%,respectively.Conclusion： Compound Fufangteng mixture has an obvious mobilizing effete on murine peripheral bloods tem cells.The effect of Fufangteng mixture combined with G-CSF is better than single usage of compound Fufangteng mixture or G-CSF.
