摘要
以不同质量浓度的铅(10、20、40、80mg/L)作用于Wish细胞3、6、12、24h,探索了铅对细胞急性死亡的影响。结果表明,10mg/L的铅使Wish细胞出现;了以 环死为特征的细胞死亡铅导致细胞死亡可通过2条不同途径:一个途径为一定剂量的铅通过启动细胞内的凋亡程序,出现以细胞体积缩小、胞质浓缩、DNA琼脂糖电泳呈梯状图像、流式细胞仪检测出现明显凋亡峰为特征的细胞凋亡;另一个途径是在铅质量浓度过大时。
This study was undertaken to characterize the acute cell death in Wish cells adding with different concentrations(10,20,40,80 mg/L) of lead following 3 24 h exposures. The different fate of Wish cell was determined by different concentration of lead .No obvious toxicity,maximal apoptotic peak ,necrotic cells were observed in Wish cells after treated with 10,40,80 mg/L lead ,respectively. There were two pathways of cell death in lead toxicity. One pathway was through triggering cell death programme that causes the apoptosis. The characteristic changes of apoptosis included the small cell volume, shrinkage with condensation of cytoplasm ,“ladder” fragment in DNA gel electrophoresis, and an apoptosis peak in flow cytometry(FCM) determination. The other pathway was an unplanned process that causes the necrosis. The characteristic features of necrosis involved the swollen cells disrupted plasma membrane, smear DNA gel electrophoresis, large cell death ratio in trypan blue examination while small number apoptotic cells in FCM measurement. The mechanisms of cell death might relate to lipid peroxidation, the metabolic disturbance of intracellular calcium, the alteration of p53 gene expression.
