5个葡萄microRNAs及其靶基因在冬芽二次成花过程中的表达 被引量:10

Spatiotemporal Expression of Five microRNAs and Their Target Genes during Flower Development of Treated Winter Buds of Grapevine in Growing Season

摘要 试验以‘藤稔’葡萄为材料,利用荧光定量PCR技术检测5个葡萄miRNAs(vv-miR160a、vv-miR171a、vv-miR159、vv-miR164c和vv-miR167c)及其靶基因在不同摘心处理的冬芽发育过程中的动态表达特征,以探讨mi-croRNAs调控花发育的分子机理。结果显示:5条miRNAs及其靶基因在二次成花的冬芽中的表达发生了明显变化。其中,vv-miR160a、vv-miR171a和vv-miR159表达水平明显增强,在花序中有最高表达;vv-miR164c和vv-miR167c的表达水平明显减弱,在花序中的表达最低或较低,而这些vv-miRNAs在对照组中表达水平均无明显变化。研究表明,上述miRNAs参与了葡萄冬芽的二次花的发育;从miRNAs与其靶基因的表达呈现的消长变化趋势看,这些vv-miRNAs通过负调控其靶基因表达而起作用。 Quantitative RT-PCR was employed to analyze the expression of five important mieroRNAs (miRNAs), namely vv-miR160a, vwmiR171a, vv-miR159, vv-miR164c and vv-miR167e, and their target genes in winter buds growing on grapevine ev. Fujiminori's shoots with treatments of cut-back at various times. Results showed that the various treatments have different effects on the expression of miRNAs and their target genes,of which vv-miR160a, vv-miR171a and vv-miR159 showed distinct increases in expres- sion in the winter buds differentiating to flower buds, and they showed the highest express level in inflores- cences,while vv-miR164c and vv-miR167c exhibited obviously decreased expression and were lowly ex- pressed in inflorescences. All the five miRNAs showed no distinct variation in the winter buds of control plants,indicating these miRNAs are involved in the second flower development of winter buds. The inverse expression trends between miRNAs and their target genes suggest that these miRNAs negatively regulate their target gene expression.
