人参皂甙Rd联合远程预处理对脑缺血大鼠的脑保护效应 被引量:2

Neuroprotective Effect of Combined Ginsenoside Rd with Remote Preconditioning after Focal Cerebral Ischemia in Rats

摘要 目的比较单独或联合应用人参皂甙Rd及远程预处理的脑保护效应,探寻安全有效的脑保护方法。方法 40只雄性SD大鼠随机均分为4组(n=10/组):①对照组(C组),腹腔注射生理盐水1h后制备大脑中动脉栓塞-再灌注(middle cerebral artery occlusion-reperfusion,MCAO)模型;②人参皂甙Rd组(Rd组),腹腔注射人参皂甙Rd30mg/kg1h后行MCAO;③远程预处理(remote preconditioning,RPC)组,远程预处理1h后行MCAO;④人参皂甙Rd联合远程预处理组(Rd+RPC组),远程预处理前1h腹腔注射人参皂甙Rd30mg/kg,远程预处理1h后行MCAO。分别在再灌注24h、72h行神经功能评分(neurological behavior scores,NBS),72h评分后取大脑行2,3,5-氯化三苯四唑(triphenyltetrazolium chloride,TTC)染色测量梗死容积百分比。结果再灌注后24h和72h,各处理组的NBS明显高于对照组(P<0.01),RPC组的NBS明显高于Rd组,而Rd+RPC组NBS又明显高于Rd组和RPC组(P<0.05)。各处理组脑梗死容积百分比明显小于对照组(P<0.01),RPC组也明显小于Rd组(P<0.01),而Rd+RPC组又明显小于Rd组和RPC组(P<0.05)。结论人参皂甙Rd及远程缺血预处理均具有明显的脑保护效应,且联合两种方法表现出更强的脑保护协同效应。 Objective To investigate the neuroprotective effect of the combined ginsenoside Rd with remote preconditioning in rats and seek the safe and effective neuroprotective method.Methods Forty male SD rats were randomly assigned to four groups(n=10 each).In the control group(group C),the rats were intraperitoneally injected with saline 1 hour before middle cerebral artery occlusion-reperfusion(MCAO) was performed.In the ginsenoside Rd group(group Rd),the rats were intraperitoneally injected with ginsenoside Rd(30 mg/kg) 1 hour before MCAO.In remote preconditioning group(group RPC),the rats received remote preconditioning(RPC) 1 hour before MCAO.In combined ginsenoside Rd with remote preconditioning group(group Rd+RPC),the rats were intraperitoneally injected with ginsenoside Rd(30 mg/kg) 1 hour before RPC and MCAO was performed 1 hour after RPC.The neurological behavior scores(NBS) were assessed at 24 and 72 hour after reperfusion respectively,and the percentage of infarct volume was determined using 2,3,5-triphenyltetrazolium chloride(TTC) staining at 72 hours after reperfusion.Results The NBS in three preconditioning groups were better than that in group C(P〈0.01),in group RPC was better than in group Rd,and in group Rd+RPC was better than in groups Rd and RPC respectively(P〈0.05).The percentage of infarct volume in three preconditioning groups were significantly lower than that in group C(P〈0.01),in group RPC was lower than in group Rd,and in group Rd+RPC was lower than in groups Rd and RPC respectively(P〈0.05).Conclusion Ginsenoside Rd and remote preconditioning have significantly neuroprotective effects in rats,and their combination exhibits synergistic protective effect.
