摘要
以亚洲百合Elite中外层鳞片为外植体,研究了光暗不同培养条件下,苄基腺嘌呤(benzylaminopurine,BA)、萘乙酸(1-naphthlcetic acid,NAA)、蔗糖、活性炭(activated charcoal,AC)、水杨酸(salicylic acid,SA)、多效唑(paclobutrazol,PP333)对试管鳞茎诱导效果及诱导途径的影响。结果表明:小鳞茎共有2种发生途径,1种是从外植体上直接诱导形成小鳞茎,第2种是外植体经诱导产生不定芽,40d后经不定芽基部膨大形成小鳞茎。光培养下小鳞茎的增殖系数高于暗培养,但小鳞茎直径小于暗培养。单因素试验中,蔗糖、AC及PP333均可促进小鳞茎的膨大;AC可以促进小鳞茎品质的提高,在含0.5g.L-1AC的培养基中,诱导出的小鳞茎抱合较紧实。SA可以促进小鳞茎繁殖系数的提高,SA浓度为0.5μmol.L-1时,小鳞茎诱导率高达96%。多因素试验结果表明,培养基MS+0.5mg.L-1BA+0.6mg.L-1NAA+0.5g.L-1AC+0.5μmol.L-1SA+5mg.L-1PP333+70g.L-1蔗糖最适合Elite试管鳞茎的生长发育。
Scales of Lilium Asiatic hybrid 'Elite' were used to study the effects of BA,NAA,sucrose,activated charcoal,salicylic acid and paclobutrazol on bulblets inducement in light or darkness culture.The results showed there were two bulblets inducement pathways.First,the bulblets were induced directly from scales.Second,buds formed in light culture and inflated into bulblets after 40 days.The multiplication of bulblets in light was higher than in dark culture.However,the diameter of bulblets induced in light was smaller than the dark.Sucrose,AC or paclobutrazol all could expand the bulblets.AC could also improve the bulblet quality.When 0.5g·L-1 AC was added to the medium,the cohesion of bulblets scale was more tightly and closely.SA improved the proliferation of bulblets.0.5 μmol·L-1 SA could increase the rate of bulblets formation,reaching to 96%.The best medium for the bulblets formation and growth in vitro was MS+0.5mg·L-1 BA+0.6mg·L-1 NAA+0.5g·L-1 AC+0.5μmol·L-1 SA+5mg·L-1 Paclobutrazol+70g·L-1 sucrose.
