致敏性杂质“高杂1”和“高杂2”在HPLC中的色谱行为不同。在溶剂系统(1)中,“高杂1”可分为H·I·1(1)、H·I·(2)、H·I·(3)和H·I·1(4)四个组分;“高杂2”在溶剂系统(2)中亦可分为 H·I·2(1)和 H·I·2(2)两部分。除H·I·1(4)具有非特异释放组织胺的功能外,其它各组分均可引发链霉素速发型过敏反应。 “高杂1”是商品链霉素中主要致敏性杂质。“高杂1”的含量和商品链霉素引发过敏反应的能力呈正相关。
The chromatographic behaviour of allergic impurities in Streptomycin, H.I.1 and H.I.2 is different from each other. Using mobile phase No. 1, H. I. 1. could be further fractionated into four peaks, which were named, H.I.1.(l), H.I.I (2), H. I. 1(3), and H. I. 1. (4) respectively. While using mobile phase No.2, H. I.2could also be fractionated into two fractions, which were named H. I.2.(1) and H. 1.2.(2) respectively.
Results showed that all fractions except the H. I. 1. (4) which is a non-specific histamine liberator, elicited immediate streptomycin anaphylaxes.
The H.I.I, was confirmed to be the main allergen in commercial streptomycin, and the content of which in the antibiotic is directly related to the anaphlactic action.
