汉防己甲素联合紫杉醇对胃癌细胞增殖和凋亡的影响 被引量:3

摘要 目的:研究汉防己甲素(tetrandrine,TET)联合化疗药物紫杉醇(paclitaxel,PTX)对人胃癌细胞增殖和凋亡的影响。方法:MKN-45细胞常规培养于RPMI-1640培养液中,细胞长至亚单层后加TET和(或)PTX,连续培养48 h。四甲基偶氮唑盐(MTT)比色法分析细胞增殖情况,DAPI染色和流式细胞仪检测细胞凋亡情况。结果:TET和PTX单药或联合均呈剂量依赖性地抑制MKN-45细胞增殖,两药联合时IC50较两药单用时显著降低(P<0.05)。DAPI染色和流式细胞仪显示TET和PTX联合用药组细胞凋亡率显著高于单药组(P<0.05)。结论:TET联合PTX对胃癌细胞的增殖抑制及凋亡诱导有良好的协同效应。 Objective:To investigate the effects of tetrandrine(TET) in combination with paclitaxel(PTX) on the proliferation and apoptosis in human gastric cancer cell line MKN-45.Methods: MKN-45 cells were routinely cultured in RPMI-1640 medium supplemented with 10% heat-inactivated fetal calf serum.Subconfluent cell cultures were treated with either TET or PTX,or TET in combination with PTX for 48 h.Cell growth and proliferation of MKN-45 were analyzed with MTT assay.DAPI staining and flow cytometric analysis were used to detect the apoptosis of MKN-45.Results: Both TET and PTX alone or the combined treatment inhibited MKN-45 cell proliferation in a dose-dependent manner.The 50% inhibitory concentration(IC50) for TET and PTX were significantly decreased when MKN-45 cells were treated simultaneously with TET plus PTX compared with the two drugs applied alone(P 0.05).DAPI staining and flow cytometric analysis showed that the apoptosis rates in the cells treated with TET and PTX were much greater than TET or PTX applied singly(P 0.05).Conclusion: TET and PTX exerted a synergistic effect on the inhibition of proliferation and the induction of apoptosis in gastric cancer cells.
