摘要
目的观察姜黄素对人结肠癌HCT-116细胞的生长抑制作用。方法以不同浓度(10-80μmol/L)的姜黄素作用于体外培养的HCT-116细胞48h,利用光镜观察各实验组癌细胞的密度与形态改变;应用MTT方法检测细胞的生长抑制率。结果经过姜黄素48h的处理后,HCT-116细胞出现了明显的生长抑制现象,随着姜黄素剂量的增加,贴壁细胞数量减少,死亡脱落细胞增多。20μmol/L、40μmol/L和80μmol/L姜黄素组与阴性对照组比较均有显著性差异(P<0.05)。结论姜黄素能显著抑制HCT-116细胞的生长,并呈现剂量依赖关系。
Objective To observe the effect of curcumin on the growth and proliferation of human colon cancer cell line HCT-116 in vitro.Methods The HCT-116 cells were treated with 10-80μmol/L curcumin for 48 hours respectively.The morphological changes of HCT-116 cells were observed with inverted microscope,The effects of curcumin on the growth of HCT-116 cells were detected by MTT assay.Results MTT method revealed that curcumin could inhibit the growth and proliferation of HCT-116 cells in a dose-dependent manner.Except low dose(10μmol/L)group,other dose curcumin treated group(20 μmol/L、40μmol/L、80μmol/L)had significant difference compared with negative control group(P〈0.05).And the growth inhibit rates of these curcumin groups had significant difference compared with the control group(P〈0.05).Conclusion Curcumin could inhibit the growth and proliferation of HCT-116 cells in a dose-dependent manner in vitro.
